When DMUs were introduced to the service Glasgow St Enoch station was used. When it closed in June 1966 Central station was used as the Glasgow terminus.
Glasgow - Kilmarnock was part of the G&SW route from Glasgow Central to Carlisle so DMUs on that service would cover the same route.
DMUs used would normally be Hamilton sets until it closed, along with Ayr and Corkerhill units.

A Met-Camm DMU seen at what is thought to be around the village of Shillford (or Shilford) between Barrhead and Dunlop in 1966. Derby Suburban Set 387 brakes for Kilmaurs station on a northbound service to Glasgow Central. The station had reopened that day and was served by Glasgow - Kilmarnock local DMU services. Auchinleck station also re-opened the same day, generally served by Glasgow - Kilmarnock - Carlisle loco hauled services. Thus a trip between the two new stations at that time required a change at Kilmarnock. The new platform visible in the distance seems to be on the former Glasgow bound formation. Derby Suburban Set 387 approaches Kilmaurs station from the south on a Kilmarnock - Glasgow working. The section from Kilmarnock to Barrhead was singled circa 1973/4 (save for a passing loop at Lugton) and the former double track formation is evident here. Kilmaurs was closed in 1966 but the station had re-opened that day under the auspices of Strathclyde PTE. Frequencies were subsequently increased to half hourly many years later in 2009 with the construction of a 5.5 mile dynamic loop (further north) between Lugton and Stewarton. 101321 (presumably 53146 closest) at Dunlop on the 12.45 Glasgow Central - Kilmarnock, 23-11-1987. A futile attempt to take a shot in poor light. This looks like the train's heading into the sunset - actually it was the middle of a cold November day. The sun glints on the rails ahead. This was once double track. The area on the right became totally overgrown. To the left (the old goods yard) is now housing, with an obscenely high fence totally hemming in the platform. In 2009, the line from Stewarton to Lugton was re-doubled, and this view of Dunlop station has been totally transformed. It is now a twin track station again, and benefitting from a half hourly service between Kilmarnock and Glasgow. Gordon Thomson.

On 28th January 1988 a lorry toppled from the A736 road bridge over the G&SW line at the south end of Uplawmoor, some of its load falling onto the track as did some bridge parapet masonry derailing the leading vehicle 10:45 Kilmarnock to Glasgow Central. There were no casualties amongst the 30 passengers, the lorry driver was unhurt. It was worked by a hybrid set, Class 101 DMBS 51248 (closest, it would be withdrawn afterwards), a Class 101 TBSL (likely 59112, the only BG TBSL at Ayr) and a Class 120 DMSL. John Reddyhoff.
